Job Summary

Description

Department: Division of Gastroenterology

Physical work location: 1425 Madison Ave New York NY 10029

Name PI or Supervisor: Saurabh Mehandru

Web link to Lab: Contact: Erica Krasny

Responsibilities

The main objective of my PhD project will be to identify dysregulated immune networks triggering and sustaining inflammation in CVID patients with GI tract disorders. My work will focus on the study of human gastrointestinal biopsies. I will use a combination of single cell RNA sequencing (scRNA-seq) high dimensional flow cytometry and whole slide high-resolution multiplex imagery approaches to identify the functional alterations of immune cells within the CVID inflamed gut and/or stomach mucosa.

The project will be structured around three specific aims (described in the following section Goals/Outcomes of the Research Project) : 1) Investigating the cellular and molecular organizations of gut inflammatory lesions in CVID ; 2) Addressing the role of B cells and autoantibodies in the pathogenesis of CVID gut inflammatory lesions ; 3) Dissecting altered molecular trajectory of myeloid cells in CVID.

This PhD project will be co-supervised by Prof. Saurabh Mehandru (MD-PhD Icahn School of Medicine at Mount Sinai New-York USA) and Prof. Jérôme Martin (PhD Nantes University France). It will be part of an international pre-existing collaboration between Nantes University and the Icahn School of Medicine at Mount Sinai contributing to a translational research effort ultimately aimed at benefiting patients.

Qualifications

Expected Outcomes :

Ultimately the expectations we may have from this project are to support the development of tailored therapeutic strategies that best suit immunocompromised individuals based on a clear understanding of the immune pathways dysregulated. This work may also unravel new knowledge on how B cells T cells and myeloid cells control gut homeostasis.
