The mRNAs encoding a wildtype gene which is mutated in the patients will be delivered to the cells by transfection or to a mouse via injection. Subcellular localization in cells and the effect of the wildtype protein on the phenotype of the patient-derived cells will be mice we will first study the distribution and stability of the delivered mRNA in various tissues. Then we will use the mouse model of the disease and will study if and how wildtype mRNA rescues mutant mouse phenotype.
