DescriptionWe are seeking a highly motivated detail-oriented Senior Research Scientist with a strong background in cell and molecular biology to join our multidisciplinary research team. Our group investigates the molecular and cellular mechanisms regulating human megakaryopoiesis hematopoietic stem cell biology and microenvironmental interactions.
The ideal candidate will have extensive hands-on experience with ex vivo culture of primary human hematopoietic stem and progenitor cells strong technical expertise in multicolor flow cytometry using the Attune NxT platform and proficiency in molecular biology and genetic manipulation techniques. This position provides an opportunity to contribute directly to cutting-edge research focused on platelet production stem cell regulation cellcell interactions and translational studies in hematology.

Responsibilities- Isolate and culture primary human hematopoietic stem and progenitor cells (CD34) and other subsets from mobilized peripheral blood or bone marrow.
- Perform ex vivo differentiation assays for megakaryocyte and erythroid lineages including morphological molecular and functional analyses.
- Perform basic mouse handling for hematology-related experiments including injections treatments tissue/bone marrow harvesting and sample processing in compliance with institutional animal care and use guidelines.
- Operate maintain and calibrate the Attune NxT flow cytometer; design optimize and troubleshoot multicolor panels for complex immunophenotyping cell sorting and quantitative analysis of hematopoietic subsets including mature megakaryocytes and platelets.
- Conduct methylcellulose-based semisolid colony-forming assays (CFU CFU-Mk) and liquid culture systems for hematopoietic progenitors.
- Perform protein-based assays (protein extraction Western blotting ELISA) for signaling and pathway analyses.
- Conduct RNA and DNA extractions RT-qPCR and standard molecular biology procedures for gene expression and validation studies.
- Utilize gene-silencing and genome-editing approaches including siRNA/shRNA CRISPR/Cas9 and lentiviral transduction in primary and immortalized cells.
- Maintain accurate well-organized laboratory notebooks records and sample inventories.
- Assist in developing optimizing and standardizing protocols for cell signaling differentiation and functional assays.
- Support data analysis figure preparation and contribute to scientific manuscripts and grant applications.
Qualifications- Ph.D in sciences or related field
- Minimum five years of experience with scientific investigation.
- Must have demonstrated outstanding achievement in a field of research
- Must have contributed to a number of peer-reviewed publications inventions or the like at least some of which have had a major impact on advancing the field or discipline.
- Prior experience successfully securing external grant funding is highly desirable.
- Preferred:
- 45 years of hands-on laboratory experience in cell biology ideally involving hematopoietic stem cells or primary human cell systems.
- Basic experience with handling laboratory mice including injections euthanasia and tissue or bone marrow collection.
- Proven expertise in flow cytometry including instrument setup multicolor compensation and data analysis (e.g. FlowJo FCS Express) for panels 8 colors.
- Strong technical skills in protein and nucleic acid analysis (Western blotting qPCR ELISA cytokine stimulation assays etc.).
- Demonstrated experience with human hematopoietic stem/progenitor cell isolation culture and megakaryocyte/erythroid differentiation including work with immortalized cell lines.
- Experience with siRNA/shRNA CRISPR/Cas9 gene editing lentiviral transduction and basic molecular cloning.
- Excellent analytical abilities organizational skills and troubleshooting capabilities.
- Ability to work both independently and collaboratively within a diverse research team.
- Receptiveness to feedback and commitment to continuous technical and scientific improvement.
- Prior experience studying human megakaryopoiesis platelet biogenesis or hematopoietic stem cell signaling.
- Experience performing colony-forming assays (CFU CFU-Mk) and liquid hematopoietic culture systems.
- Experience with signaling pathway analyses (e.g. intracellular-flow cytometry cytokine or inhibitor-based stimulation).
- Experience with gene silencing approaches or viral transduction systems (lentiviral/retroviral) and CRISPR/Cas9 editing.
- Knowledge of immunofluorescence microscopy and image-based analysis for assessing cellular morphology and megakaryocyte maturation.
- Familiarity with bioinformatics tools for cytometry and gene-expression analysis (FlowJo FCS Express Prism etc.).
- Previous experience in translational hematology or stem cell therapyrelated research.
- Experience performing mouse models of hematologic disease including dosing monitoring and ex vivo analysis of harvested tissues.
