Project Description:
Dr. Pietro Fontana Assistant Member in the Center of Excellence for Pediatric Immuno-Oncology (CEPIO) and Department of Immunology is seeking passionate and highly motivated candidates for a fully funded postdoctoral fellowship in either Structural Biology or Cell Biology. The Fontanalab focuses on understanding how immune receptors recognize danger signals and initiate downstream responses with a particular emphasis on the mechanistic basis of activation and small molecule modulation.We investigate pathways involving cytosolic DNA sensors inflammasome components and adaptor proteins such as cGAS AIM2 and TLRs. Our research integrates cryo-EM cell biology and chemical biology to dissect how molecular architecture governs immune function. Recent work includes the discovery and structural characterization of novel dsDNA sensors and the identification of small molecules targeting key innate immune components Responsibilities:
Option 1: Structural Biology
Express and purify immune signaling proteins and complexes using bacterial insect and mammalian systems.
Prepare samples for structural studies by cryo-electron microscopy (cryo-EM) and/or X-ray crystallography.
Perform data processing and model building (e.g. Relion CryoSPARC Coot PyMOL).
Option 2: Cell Biology
Design and execute cell-based assays to investigate innate immune signaling pathways including interferon responses NfKb pathway and cell death.
Engineer and characterize genetic models including CRISPR/Cas9 knockouts and inducible systems in mammalian cells.
Analyze immune pathway activation using molecular biology techniques (qPCR Western blotting ELISA flow cytometry) and microscopy.
Additional training in immunology structural biology biochemistry and cell biology will be provided for highly motivated candidates.
Minimum Education and/or Training:
Successful applicants will hold a Ph.D. in cell biology structural biology or related discipline. The candidate should demonstrate an academic record of excellence and a strong interest in the growing field of structural immunology.
Special Skills Knowledge and Abilities:
Option 1: Structural Biology
Experience with protein expression purification and biochemical characterization using bacterial insect or mammalian cell culture.
Good working knowledge of single particle cryo-EM pipeline and data processing.
Prior knowledge of basic cell culture and cell-based assays
Option 2: Cell Biology
Experience with mammalian cell culture gene editing (CRISPR/Cas9) and generation of stable cell lines (lentivirus).
Strong background in cell signaling innate immunity or related areas.
Proficiency in molecular biology and immunoassays (qPCR Western blotting ELISA flow cytometry reporter assays).
Experience with fluorescence microscopy and imaging-based assays.
Excellent communication and presentation skills and contribute to preparation of high-quality manuscripts.
