The laboratory studies regulation of innate inflammation within the gastrointestinal tract with the goal of developing therapies for inflammatory bowel diseases such as Crohns disease and ulcerative colitis. In particular we study the interaction between proinflammatory and antiinflammatory factors in the regulation of innate mucosal inflammatory responses using in vivo murine colitis models and in vitro primary cell culture. Our major focus has been to understand the interrelated roles of NFkB and IL10 in regulating inflammatory gene expression. The goal of the current project will be to understand the mechanisms responsible for IL10mediated inhibition and evaluate IL10 based therapies in mouse models.

PRINCIPAL DUTIES AND RESPONSIBILITIES:

Investigate the basis for IL10mediated regulation of TLRinduced gene expression by primary murine macrophages. Techniques include retroviral and lentiviralmediated gene transduction siRNA mediated knockdown RTPCR western blotting immunoprecipitation and chromatin immunoprecipitation.
Characterize the ability of selected compounds to inhibit colitis in mouse models and perform immunophenotypic analysis of treated animals including flow cytometry of mucosal immune cells gene expression analysis and ELISA of cytokines secreted from tissue biopsies.

Critical analyze obtained data prepare data for publication and presentation and with supervision from PI prepare manuscripts for submission.
